The incident happened at around 8.45pm on Friday when a man entered Scotbet in Park Road, Ardrossan, and threatened a female member of staff with a knife and demanded money.
He made off from the shop with a four-figure sum of cash, police said.
The suspect is white, around 5ft 6in and was wearing dark coloured clothing with his face covered.
Officers said the 54-year-old worker was not injured but has been left badly shaken''.
Detective Sergeant Stuart Dougan said: I would appeal to anyone who was in the area of Park Road in Ardrossan around the time of the incident on Friday night who saw a man acting suspiciously to come forward.
I would also ask anyone with information that may assist our investigation to contact Police Scotland on 101.''
