The former St Mirren defender made his move to Ibrox from the Buddies last summer and while the rest of his team-mates struggled, the 30-year-old won plaudits for his committed displays.
He made 50 appearances under former bosses Ally McCoist, Kenny McDowall and Stuart McCall, scoring five goals.
But the arrival of Mark Warburton as manager saw the former junior player drop down the Light Blues' pecking order and he now leaves by mutual consent.
McGregor failed to feature in any of the club's six matches so far as Danny Wilson and Rob Keirnan were preferred at centre-half.
Warburton, speaking ahead of Wednesday's League Cup tie at Airdrie and before news of McGregor's departure was announced, confirmed "a couple'' of players might leave before the transfer window shuts next Tuesday.
A club spokesman said: "Darren leaves with the club's best wishes for a successful future.''
