The accident happened at around 4.55am on the A92 north of Ladybank when a white Volkswagen Up collided with a silver Vauxhall Corsa.
Police said both drivers suffered serious injuries and were taken to Ninewells Hospital in Dundee for treatment.
Sergeant Alastair Purvis said: “The A92 remains closed whilst the collision is investigated and the road is cleared and made safe for traffic again. Diversions are in place and I would like to thank motorists for their patience.
“I'm keen to hear from anyone who may have witnessed this collision or the manner of driving of either vehicle beforehand.
