People living near the river were woken as a coastguard chopper hovered above their homes, shining a powerful light onto the water and the banks.
Police haven't found anyone yet, but say they're still searching.
Inspector Mark Chivers said: “Around 2.10am we received a report of concern for a man possibly entering the River Clyde at Bothwell Road, Hamilton. Officers attended and there was no trace in the area.
“Further searches are ongoing and at this time we ask anyone who was in the area and may have witnessed anything to come forward.
“We would also be keen to hear from any motorists who may have been driving past around that time and have dashcam footage from the area.”
