Two drivers, both male and aged 23 and 48 years old, were taken to hospital, one by air ambulance to Glasgow and the other by road ambulance to Dumfries, following a two-car crash on the A74(M) near Johnstonebridge.
The incident happened around 1 o'clock on Tuesday afternoon, involving a black Seat Leon and a white Tesla.
The road was closed for several hours while police investigated, before reopening later at night.
Officers are appealing for witnesses or anyone with dashcam footage to contact Police Scotland.
Sergeant Colin Ramage said: “I’m appealing for anyone who saw what happened, or saw either vehicle before the collision, to come forward.
“I’d also ask drivers with dash-cams to review any footage of the area around the time to see if you have captured something which may assist our investigation.
“Members of the public are thanked for their patience yesterday. We appreciate this caused delays on the road network, however it was essential to ensure the safety of officers and other road users while enquiries were carried out.”
Anyone with information is urged to contact Police Scotland on 101 quoting reference 1412 of 11 November.
