Billy Mckay broke his goalscoring drought with two efforts that ensured Caley took advantage of Dundee United's slip-up on Friday night.
Inverness made one change from the side that lost narrowly at Celtic last weekend. Carl Tremarco dropped out of the starting line-up with Mckay coming in at his expense.
Hamilton made three changes from the dramatic 3-3 draw with Partick Thistle as Grant Gillespie, Martin Canning and Dougie Imrie were replaced by Stephen Hendrie, Michael Devlin and Jon Routledge.
Playing under a blazing November sun, the home side started in the ascendancy through Mckay, who capitalised on a Michael Devlin slip with a pull-back into the area. James Vincent hurdled the ball and it broke to Greg Tansey, who curled his effort just wide.
The visitors took the lead in speculative fashion as Tony Andreu lined up a free kick from 35 yards, toe-poked it towards goal and hit the inside of the post before it went beyond Dean Brill.
However, Caley Thistle would only be behind for 10 minutes as last season's leading marksman Mckay got on the end of Marley Watkins' cross and steered his effort beyond Michael McGovern.
Referee Willie Collum had a situation to diffuse on the half-hour as Graeme Shinnie and Jon Routledge went to square up to each other. Yellow cards weren't required, however, as the pair shook hands amicably after a quick word from Collum.
Inverness came close to taking the lead five minutes before the break with Mckay looking for his second, latching on to Watkins' slip and angling a right-footed effort just beyond McGovern's far post.
In a fairly tepid start to the second half, a Watkins burst down the right flank was a rare glimpse of quality. He did well to hold off Jesus Garcia Tena before steering a ball to the back post aiming for Vincent but unfortunately it was too close to McGovern who claimed well.
The next delivery into the box was altogether more potent, Tansey swinging a corner in that Gary Warren planted a bullet header on and left McGovern with no chance.
Inverness were in the ascendancy and found particular joy on the counter attack, Watkins and Shinnie having success. Shinnie tried his luck from 25 yards on one break, stinging the fingertips of McGovern.
The goals soon started to flow in the Highland Capital, first Mckay burying the rebound when Watkins had a shot saved, before Andreu got his second with a shot that squirmed under Brill.
Not to be denied his goal, Vincent latched on to a Watkins through ball and slid the ball round McGovern, for his first goal since the start of last season.
Dougie Imrie, returning to his former stomping ground, had the hosts worried with five minutes to go when he cut inside but unfortunately for the Accies his effort fizzed wide.
